dc.description.abstract | Mezenhimālajām stromālām šūnām jeb meznhimālajām cilmes šūnām (MCŠ) piemīt pašatjaunošanās spējas un tās var diferencēties vairāku līniju šūnās. Paaugstināts aktīvo skābekļa formu daudzums var ietekmēt MCŠ pašatjaunošanās spējas, diferenciācijas kapacitāti un proliferāciju. Spirulīna satur vairākus spēcīgus, pierādītus antioksidantus kā fikocianīns, fikocianobilīns, polisaharīdi, β-karotīns, C un E vitamīni, taču tās antioksidatīvais potenciāls ir atkarīgs no augšanas apstākļiem, ekstrakta sagatavošanas, uzņemšanas veida un citiem faktoriem. Darba mērķis ir novērtēt, kāda ir Latvijā audzētas un aukstumā žāvētas aļģes spirulīna ietekme uz oksidatīvajam stresam pakļautām mezenhimālām stromālām šūnām. Pēc ūdeņraža peroksīda izraisīta oksidatīvā stresa in vitro, MCŠ, kurām iepriekš pievienots 200µM spirulīnas ekstrakts, vārpstveida šūnu skaits bija par 19,7% lielāks salīdzinot ar kontroles grupu. Pievienojot 100µM spirulīnas ekstraktu netika novērotas statistiski nozīmīgas atšķirības starp grupām. Tiek uzskatīts, ka vārpstveida šūnu skaits korelē ar MCŠ dzīvspēju. | lv-LV |
dc.description.abstract | Annotation Mesenchymal stromal cells or mesenchymal stem cells (MSC) possess a self-renewal ability and can differentiate into cells of multiple lineages. In MSCs, excess reactive oxygen species can impair self-renewal, differentiation capacity, and proliferation. Spirulina contains several powerful, proven antioxidants such as phycocyanin, phycocyanobilin, polysaccharides, β-carotene, and vitamins C and E. However, its antioxidant potential depends on growth conditions, extract preparation, method of intake, and other factors. The goal of this study is to evaluate the effect of Spirulina, grown in Latvia and freeze-dried, on mesenchymal stromal cells subjected to oxidative stress. After oxidative stress induced by hydrogen peroxide in vitro, MSCs to which 200µM of Spirulina extract had been previously added showed a 19.7% higher count of spindle-shaped cells compared to the control group. Adding 100µM of Spirulina extract did not show statistically significant differences between groups. It is considered that the number of spindle-shaped cells correlates with the viability of MSCs. | en-UK |
