Amorfās cietās dispersijas (ASD) uzlabo slikti šķīstošo zāļu biopieejamību, iekļaujot aktīvo polimēru matricā, kas visbiežāk sastāv no diviem vai vairākiem polimēriem, kas ir savstarpēji papildinoši. Viena no pētāmajām kombinācijām ir Soluplus un hipromelozes acetāta sukcināts (HPMC-AS LF). Precīza polimēra kvantitatīvā noteikšana ir svarīga zāļu formas šķīdībai, stabilitātei, pareizajai aktīvas vielas un palīgvielu izdalīšanā. Iepriekšējās kvantitatīvās noteikšanas metodēs tika izmantota SEC-UV Soluplus un augstas veiktspējas šķidruma hromatogrāfija ar iztvaikošanas gaismas izkliedes detektoru (HPLC-ELSD) HPMC-AS LF noteikšanai. Šajā pētījumā tika izstrādāta alternatīva metode HPMC-AS LF kvantitatīvai noteikšanai, izmantojot reflux metodi, kuras rezultātā veidojas HPMC-AS LF polimēra un metāla (hroma) komplekss, var ļaut netiešā veidā kvantitatīvi noteikt polimēra koncentrāciju šķīdumā. HPMC-AS LF kompleksu veidošanās ar hromu tika apstiprināta, izmantojot FTIR analīzi, vizuālo pārbaudi, kā arī optisko mikroskopiju. Cr+ jonu koncentrācijas samazināšanās šķīdumā izgulsnējušo kompleksu dēļ tika konstatēta, izmērot atlikušos hroma jonus saturošā šķīduma UV absorbciju pēc kompleksa veidošanās procesa pie 242 nm viļņa garuma, izmantojot pirmo atvasinājumu analīzi, kas ļāva netieši kvantificēt HPMC-AS LF. Lai gan metodei joprojām ir nepieciešama optimizācija, pašreizējās metodes atkārtojamība ar piecu stundu reflux laiku ir 1,79%. HPMC-AS LF tika netieši kvantitatīvi noteikts pēc kompleksēšanas ar hromu, mērot šķīdumā atlikušā hroma UV absorbciju. Lai gan metodes precizitāte un precizitāte vēl ir jāuzlabo, tās rentabilitāte un pieejamība attaisno ieviešanu, novēršot vajadzību pēc specializētiem HPLC detektoriem un kolonnām, kas nepieciešamas parastajai polimēru kvantitatīvai noteikšanai.
Amorphous solid dispersions (ASDs) enhance the bioavailability of poorly soluble drugs by incorporating the API in a polymeric matrix, sometimes consisting of two and more polymers, which are complementary. One of the combinations under investigation is Soluplus and hypromellose acetate succinate (HPMC-AS LF) polymers. Precise polymer quantification is crucial for the solubility, stability, API and excipients release of the final dosage form. While previous quantification methods employed SEC-UV for Soluplus and high-performance liquid chromatography with evaporative light-scattering detection (HPLC-ELSD) for HPMC-AS LF, in this study an alternative method for the quantification of HPMC-AS LF was investigated through a reflux-based metal-polymer framework (MPF) approach which may allow the indirect quantification of HPMC-AS LF. The complexation of HPMC-AS LF with chromium was confirmed via FTIR, visual inspection, as well as optical microscopy. The decrease in Cr+ ions concentration in solution due to the precipitating complexes was detected by measuring the UV absorbance of the solution containing the remaining chromium ions after the MPF formation process at 242 nm wavelength using first derivative analysis, which allowed for the indirect quantification of HPMC-AS LF. While the method still requires optimization, repeatability of the current method with a reflux time of five hours demonstrates an R.S.D.% of 1.79%. HPMC-AS LF was indirectly quantified after complexation with chromium by measuring the UV-absorbance of leftover chromium in solution. While the accuracy and precision of the method remains to be improved, its cost-effectiveness and accessibility justify implementation, eliminating the need for specialized HPLC detectors and columns required for conventional polymer quantification.
