Šī darba mērķis bija izstrādāt un novērtēt inovatīvu kolagēna-akrila hidrogēlu, kas paredzēts mezenhimālo cilmes šūnu (MSC) kultivācijai un izmantošanai reģeneratīvās medicīnas kontekstā. Tika sintezēts strukturāli stabils un sorbcētspējīgs hidrogēls, uz kura tika uzsētas MSC no donora nabassaites. Darba gaitā tika analizēta šūnu dzīvotspēja, proliferācija, MSC spēja diferencēties endotēlijas virzienā, šūnu migrācija hidrogēlā uz iekaisuma signāliem, 3D šūnu infiltrācija hidrogēlā un fenotipa saglabāšana pēc kultivācijas uz hidrogēla. Rezultāti liecina par augstu MSC dzīvotspēju un funkcionālo potenciālu uz materiāla, kā arī strukturālu atbilstību terapeitiskiem pielietojumiem. Materiāls uzskatāms par daudzsološu pielietošanai reģeneratīvajā medicīnā, t.sk. brūču terapijā.
The aim of this work was to develop and evaluate an innovative collagen-acrylic hydrogel intended for the cultivation of mesenchymal stem cells (MSCs) and its application in regenerative medicine. A structurally stable and absorbent hydrogel was synthesized, on which MSCs from a donor umbilical cord were cultured. The study analyzed cell viability, proliferation, the ability of MSCs to differentiate into the endothelial lineage, cell migration on the hydrogel in response to inflammatory signals, 3D cell infiltration in the hydrogel, and the maintenance of phenotype after cultivation on the hydrogel. The results indicate high MSC viability and functional potential on the material, as well as structural suitability for therapeutic applications. The material is considered promising for regenerative medicine, including wound healing therapy.
