Kopsavilkums
Mērķis. Ievākt paraugus no 44 RSU ēkās esošiem ūdens aparātu krāniem, identificēt izaugušo koloniju mikroorganismus un salīdzināt pārstāvju atšķirības starp krāna ārējo un iekšjo virsmu.
Materiāli un metodes. Tika paņemti 44 nomazgājumi no ūdens aparatu krānu ārējas un iekšējas virsmas. Kopumā tie bija 22 ūdens aparāti, kuri atradās 4 Rīga Stradiņš University ēkās. Nomazgājumu materiāls tika uzsēts uz asins un Sabouraud agariem. Izaugušas uz plates kolonijas tika saskaitītas un katra atsevišķi nokrāsota pēc Grama metodes vai ar metilēnzilo un mikroskopētas 100x palielinājumā. Randomizēti izvēlētās kolonijas tika pavairotas un identificētas ar automated microbiology system VITEK2 programmu.
Rezultāti. Uz asins agariem izaugušo koloniju skaita reference sastādija no 0 līdz 10000 KVV/ml. Uz Sabouraud agariem izaugušo koloniju skaita reference sastādija no 0 līdz 5000 KVV/ml. No ūdens aparātu krānu iekšējās virsmas paņemtiem nomazgājumiem izauga kolonijas uz visām 22 asins agara barotnēm un uz 12 no 22 Sabouraud agara barotnēm. No ūdens aparātu krānu ārējās virsmas paņemtiem nomazgājumiem izauga kolonijas uz visām 22 asins agara barotnēm un uz 20 no 22 Sabouraud agara barotnēm. Iekšējās un ārējās virsmas KVV/ml attiecība sastāda uz asins agara barotnēm (KVV/ml); Sabouraud barotnēm (KVV/ml); to var skaidrot ar ārējās virsmas lielāku kontaminēšanas risku ar roku un priekšmetu mikroorganismiem. Visbiežāk identificētas grampozitīvās baktērijas pieder Staphylococcaceae (11 no 60 identificētām baktērijām) un Micrococcaceae (14/60) family. Visbiežāk identificētas gramnegatīvās baktērijas pieder Sphingomonadaceae (9/60) un Moraxellaceae (8/60) family. Visbiežāk identificētais mikroorganisms izrādijās gramnegatīva strictly aerobic baktērija Sphingomonas paucimobilis.
Secinājums. Iepazīstoties ar konstatēto mikroorganismu uzbūvi, īpašībām, patoģenitātes faktoriem un iespējamo ietekmi uz cilvēka veselību secināts ka lielākā daļa ir normālā cilvēka mikrobiota pārstāvji un kaitējumu veselībai nenes.
Objectives. Water dispensers are used to provide fresh water for RSU workers and students. The frequent use of water dispenser taps increases the risk of microbial colonization, which may lead to water contamination with potentially pathogenic microorganisms. The aim of the present study was to count and identify microorganisms colonizing the inner and outer surface of water dispensers. Materials and Methods. For this study, 44 samples were taken from the outer and inner surface of 22 water dispenser taps. The samples were cultivated on Blood and Sabouraud agars. All of the colonies were counted and stained using Gram or methylene blue stains. VITEK2 system was used for identification. Results.After inoculation of the samples, which were taken from the inner surface of water dispenser taps, colonies formed on 22 out of 22 Blood agars and 12/22 Sabouraud agars. Colonies formed on 22/22 Blood agars and 20/22 Sabouraud agars in the inoculated samples from the outer surface of the taps. The average count of microorganisms in the samples of the outer surface was 976 for Blood agars and for Sabouraud agars. The average count of microorganisms from the inner surface was 612 for Blood agars and 19 for Sabouraud agars. Most of the Gram-positive bacteria that were identified belonged to Staphylococcaceae and Micrococcaceae families. Most of the Gram-negative bacteria that were identified belonged to Sphingomonadaceae and Moraxellaceae families. The most common identified bacterium was Sphingomonas paucimobilis. Conclusions.The major part of microorganisms found in the samples taken from the surface of water dispenser taps was representatives of normal human microbiota that were not associated with infectious diseases in a healthy human. Sphingomonas paucimobilis was the most common identified bacterium.The outer surface of the water dispenser taps showed more notable contamination that the inner surface.
