Parkinsona slimība ir progresējoša neirodeģeneratīva kustību traucējumu slimība, kuras agrīna diagnostika joprojām ir būtisks izaicinājums. Slimības prodroma stadijā raksturīgās pārmaiņas smadzenēs sākas krietni pirms klīnisko simptomu parādīšanās, tādēļ agrīnas diagnostikas uzlabošanai arvien lielāka nozīme tiek piešķirta attēldiagnostikai un biomarķieru meklējumiem. Šajā pētījumā padziļināti analizētas substantia nigra un tajā esošā neiromelanīna tilpuma izmaiņas Parkinsona slimības pacientiem, izmantojot augstas izšķirtspējas magnētiskās rezonanses neiromelanīnam jutīgu sekvenci un pusautomātisku attēlu apstrādi. Novērota būtiska neiromelanīna signāla samazināšanās Parkinsona slimības pacientu grupā, kas korelē ar motorisko simptomu izteiktību. Pētījuma rezultāti liecina, ka neiromelanīna kvantitatīva novērtēšana var kalpot kā daudzsološs biomarķieris agrīnai diagnostikai un slimības gaitas uzraudzībai, tādējādi uzlabojot pacienta dzīves kvalitāti un ārstēšanas iespējas.
Parkinson’s disease is a progressive neurodegenerative movement disorder, and its early diagnosis remains a major challenge. During the prodromal phase, characteristic brain changes begin long before the onset of clinical symptoms. Therefore, increasing attention is being paid to imaging diagnostics and biomarker discovery to improve early detection. This study provides an in-depth analysis of volume changes in the substantia nigra and its neuromelanin content in Parkinson’s disease patients, using a high-resolution neuromelanin-sensitive MRI sequence and semi-automated image analysis. A significant reduction in the neuromelanin signal was observed in the Parkinson’s disease group, correlating with the severity of motor symptoms. The results suggest that quantitative evaluation of neuromelanin may serve as a promising biomarker for early diagnosis and disease monitoring, ultimately improving patient quality of life and treatment outcomes.
